TIRF image of 0.3 micron yellow/green spheres excited by a 488nm laser line. [00:28] - Spheres attached to the coverslip are constantly visible whereas other spheres are seen drifting in and out of the field of view via Brownian motion. This demonstrates the superb ~100nm axial resolution available with TIRF microscopy.
